I have an all-in-one printer/scanner. It is connected to the desktop
computer via a USB2 cable. Now I want to do some scanning downstairs. Can I
pair a bluetooth on the computer to another one on the printer, trying to
make it a wireless network?

Nope. The printer is a USB client not a host so it can't drive a USB dongle
or anything else.
You need some sort of USB print server device to connect to the printer and
then connect this to a wireless network (WiFi or bluetooth, but I would
recommend WiFi if you need to go through walls/floors as bluetooth generally
isn't strong enough).
Some cheap routers have a USB print server built in.

in the market there's a few models of printers you can choose from.
1. Wifi
2. Ethernet
3. Bluetooth
4. USB
5. LPT
So when you buy your printer you need to check what you need and choose accordingly... or if you wan, you can buy a printer server like what most Networking companies like Linksys, Dlink, Buffalo, or even HP. has...
question... why is your printer so far away from your computer?
the problem now is that you need to make sure that the scanner downstairs is bluetooth compatible and make sure that the software that is (or will be used) is also compatible with the devices willing to be used
